MyoCult™ Differentiation Kit (Human)

Medium for the differentiation of human skeletal muscle progenitor cells (myoblasts) into myotubes
概要
MyoCult™ Differentiation Kit (Human) includes MyoCult™ Differentiation Medium and Animal Component-Free (ACF) Cell Attachment Substrate. MyoCult™ Differentiation Medium is a serum-based medium specifically formulated for the in vitro differentiation of human skeletal muscle progenitor cells (myoblasts) into myotubes. MyoCult™ Differentiation Medium is designed for use with the ACF Cell Attachment Substrate, which has been specifically tested for its ability to support optimal adherence and myotube morphology. This medium is suitable for the differentiation of myogenic progenitor cells previously expanded in serum-containing or serum-free media (e.g. using the MyoCult™-SF Expansion Kit #05980).
Advantages
• Supports differentiation of myogenic progenitor cells to multinucleated myotubes with strong expression of myosin heavy chain and myogenin markers
• Rigorous raw material screening and quality control minimize lot-to-lot variability
• Attachment substrate is included to support optimal adherence and enhanced myotube morphology
Components
  • MyoCult™ Differentiation Basal Medium (Human), 190 mL
  • MyoCult™ Differentiation 20X Supplement (Human), 10 mL
  • ACF Cell Attachment Substrate, 1 mL

Figure 1. MyoCult™ Differentiation Medium Induces Robust Differentiation of Myotubes from Human Myoblasts

(A) Myogenic progenitor cells expanded in MyoCult expansion medium expressed MyoD (green) and desmin (red). (B) Robust differentiation of myoblasts (shown in (A)) to multinucleated myotubes expressing myosin heavy chain (MyHC+) was achieved following culture in MyoCult™ Differentiation Medium for as little as 5 days. (C) A fusion index showing approximately 75% of total nuclei were found within myotubes expressing MyHC.
